چکیده

Objective: Brown fat activation attenuates atherosclerosis development by accelerating triglyceride-rich lipoprotein turnover and/or stimulation of reverse cholesterol transport via the SRB1 (scavenger receptor class B type 1). The aim this study was to investigate specific role hepatic in atheroprotective properties brown activation. Approach and Results: APOE*3-Leiden.CETP mice, a well-established model human-like metabolism atherosclerosis, were treated with vehicle or adenoassociated virus serotype 8-short hairpin RNA, which decreased protein levels 40% 55%. After 2 weeks, mice without knockdown β3-adrenergic agonist CL316 243 activate for 4 weeks determine HDL (high-density lipoprotein) catabolism 9 evaluate atherosclerosis. Surprisingly, additively improved beneficial effects agonism on development. In fact, per se not only increased HDL-cholesterol but also reduced plasma triglyceride non-HDL-cholesterol levels, thus explaining reduction Mechanistic studies indicated that is due lipolytic processing uptake VLDL (very low density facilitating VLDL-surface transfer HDL. Conclusions: Hepatic mouse an intact ApoE (apolipoprotein E)-LDLR (low receptor) clearance pathway, relevant human metabolism, effect development, explained pleiotropic VLDL. could be effective strategy treat cardiovascular disease subjects impaired function.

Inhibition of hepatic scavenger receptor-class B type I by RNA interference decreases atherosclerosis in rabbits

OBJECTIVE Scavenger receptor-class B type I (SR-BI), the receptor for HDL-cholesterol, plays a key role in HDL metabolism, whole body cholesterol homeostasis, and reverse cholesterol transport. We investigated the in vivo impact of hepatic SR-BI inhibition on lipoprotein metabolism and the development of atherosclerosis employing RNA interference. METHODS Small hairpin RNA plasmid specific fo...

Hyperglycemia suppresses hepatic scavenger receptor class B type I expression.

Hyperglycemia is a major risk factor for atherosclerotic disease. Hepatic scavenger receptor class B type I (SR-BI) binds HDL particles that mediate reverse cholesterol transport and thus lowers the risk of atherosclerosis. Here we examined glucose regulation of SR-BI gene expression in both HepG2 cells and whole animals. Results showed that hepatic SR-BI mRNA, protein, and uptake of cholestero...

Endothelial Expression of Scavenger Receptor Class B, Type I Protects against Development of Atherosclerosis in Mice

The role of scavenger receptor class B, type I (SR-BI) in endothelial cells (EC) was examined in several novel transgenic mouse models expressing SR-BI in endothelium of mice with normal C57Bl6/N, apoE-KO, or Scarb1-KO backgrounds. Mice were also created expressing SR-BI exclusively in endothelium and liver. Endothelial expression of the Tie2-Scarb1 transgene had no significant effect on plasma...

Brown fat activation reduces hypercholesterolaemia and protects from atherosclerosis development

Brown adipose tissue (BAT) combusts high amounts of fatty acids, thereby lowering plasma triglyceride levels and reducing obesity. However, the precise role of BAT in plasma cholesterol metabolism and atherosclerosis development remains unclear. Here we show that BAT activation by β3-adrenergic receptor stimulation protects from atherosclerosis in hyperlipidemic APOE*3-Leiden.CETP mice, a well-...
